1. Why this box exists

The main concern researchers raise when studying body-weight reduction is the loss of not only fat but also muscle tissue. Alongside this, a decline in energy is observed: the body shifts into conservation mode and a person feels a drop in strength.

This box is assembled around a different approach. Retatrutide is studied in the context of metabolic body-weight research. MOTS-C is a mitochondrial peptide studied for its ability to influence cellular energetics. CJC-1295 + Ipamorelin is a pair of secretory peptides researched in the direction of supporting endogenous growth hormone, which is associated with the preservation of muscle mass and tone.

The logic: fat goes — but not strength, not muscle.

2. Retatrutide — a triple metabolic agonist

The next evolutionary step after tirzepatide. Retatrutide is a synthetic 39-amino-acid peptide that engages three receptors at once: GLP-1, GIP, and the glucagon receptor (GCGR). Where tirzepatide acts on two of the three, Retatrutide recruits all three — and it is the third, the glucagon receptor, that draws particular interest from researchers, since it is linked to energy expenditure and fat oxidation.

In the study by Jastreboff A.M. et al., published in The New England Journal of Medicine (2023), participants receiving Retatrutide within a phase 2 trial showed a reduction in body weight of up to 24.2% over 48 weeks. At the study's conclusion no plateau had been reached — the curve was still trending downward [1].

3. MOTS-C — energy at the mitochondrial level

MOTS-C is a 16-amino-acid peptide encoded by mitochondrial DNA. It is one of the few known peptides produced not by the nuclear genome but by the mitochondria — the cell's "power stations."

In the study by Lee C. et al. (Cell Metabolism, 2015), MOTS-C was shown to influence glucose metabolism and the activation of the AMPK pathway — a key regulator of cellular energetics. In experimental models, increased insulin sensitivity and improved glucose handling were observed [2].

MOTS-C concentration in the body declines with age. The scientific literature discusses the link between this decline and age-related deterioration of metabolism. In the context of this box, MOTS-C is studied as a complement to Retatrutide: where the triple agonist acts at the receptor level, MOTS-C is researched at the level of the mitochondria — where cellular energy is born.

4. CJC-1295 + Ipamorelin — natural rise of growth hormone

Two peptides that work as a pair. CJC-1295 is a modified analog of growth-hormone-releasing hormone (GHRH), while Ipamorelin is a selective secretagogue acting through the ghrelin receptor. Together they are studied for their ability to stimulate the body's own growth hormone gently and physiologically.

In the study by Teichman S.L. et al. (J Clin Endocrinol Metab, 2006), CJC-1295 was shown to provide prolonged stimulation of GH and IGF-1 secretion in healthy adults [3]. Ipamorelin, unlike earlier secretagogues, is distinguished by high selectivity — in studies, minimal effect on cortisol and prolactin was observed [4].

Growth hormone is associated with the preservation of muscle mass, skin density, and overall body composition. In the context of this box, CJC-1295 + Ipamorelin is studied as support for muscle tone and body structure during metabolic changes.

5. How the components complement one another

Three components — three levels: receptors, mitochondria, growth hormone.

ComponentResearch focus
Retatrutide 20 mgMetabolic body-weight research (triple GLP-1/GIP/GCGR agonist)
MOTS-CCellular energetics: mitochondria, AMPK pathway, glucose metabolism
CJC-1295 + IpamorelinSupport of endogenous growth hormone: muscle tone, body composition

The box brings them together for researchers interested in the question: how to study body-weight reduction without the loss of muscle mass and energy.

6. Why 20 mg

Box 03 is available in two Retatrutide options: 10 mg and 20 mg. The 20 mg option is intended for longer research protocols. MOTS-C and CJC-1295 + Ipamorelin are identical in both options.
